What is a whole mount?
A whole mount is a small organism or body part placed on the slide intact, without cutting. Organisms such as Hydra or a flea, or an insect leg, are fixed, stained if needed, cleared and sealed under a cover glass.
What does H and E staining show?
Haematoxylin stains nuclei blue to purple and eosin stains cytoplasm, muscle and collagen shades of pink. The contrast shows the layers of an organ and the shape and arrangement of cells within each tissue.
How are white blood cells identified on a blood smear?
By the shape of the nucleus and the granules in the cytoplasm. Neutrophils have a lobed nucleus, eosinophils red-orange granules, basophils dark blue granules, lymphocytes a large round nucleus and monocytes a kidney-shaped nucleus.
Do zoology slides need an oil-immersion lens?
